Application Notes
Research Application(s): ELISA Detection: B27 monoclonal antibody is useful as a detection antibody in a sandwich ELISA for quantifying human IFN- gamma protein levels.1,2 Biotinylated B27 antibody should be paired with purified A35 antibody as the capture antibody, with purified recombinant human IFN- gamma as the standard.

Immunofluorescence/Flow Cytometry: B27 antibodies are useful for intracytoplasmic staining and flow cytometric analysis to identify and enumerate IFN- gamma -positive cells within mixed cell populations. Recombinant human IFN- gamma, unlabeled B27, or Rat IgG1 should be used as a specificity control.

Immunoprecipitation: The B27 antibody has been reported to be useful for immunoprecipitation.1

Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.

Characterization
To ensure acceptable performance, each batch of product is tested in a sandwich ELISA to conform to characteristics of a standard reference reagent.
Preparation and Storage
The purified (UNLB) antibody is supplied as 0.5 mg purified immunoglobulin in 1.0 mL of 100 mM borate buffered saline, pH 8.2. No preservatives or amine-containing buffer salts added. Store at 2- 8 degree C.
The fluorescein (FITC) conjugate is supplied as 100 tests in 1.0 mL PBS/NaN3. Store at 2-8 degree C.
The biotin (BIOT) conjugate is supplied as 0.5 mg labeled antibody in 1.0 mL of PBS/NaN3. Store at 2-8 degree C.
The R-phycoerythrin (R-PE) conjugate is supplied as 100 tests in 1.0 mL of PBS/NaN3 and a stabilizing agent. Store at 2-8 degree C. Do not freeze!
Reagents are stable for the period shown on the label if stored as directed.

NCBI Description

This gene encodes a member of the type II interferon family. The protein encoded is a soluble cytokine with antiviral, immunoregulatory and anti-tumor properties and is a potent activator of macrophages. Mutations in this gene are associated with aplastic anemia.

Uniprot Description

Function: Produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.

Subunit structure: Homodimer.

Subcellular location: Secreted.

Tissue specificity: Released primarily from activated T lymphocytes.

Post-translational modification: Proteolytic processing produces C-terminal heterogeneity, with proteins ending alternatively at Gly-150, Met-157 or Gly-161.

Involvement in disease: In Caucasians, genetic variation in IFNG is associated with the risk of aplastic anemia (AA) [

MIM:609135]. AA is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.

Pharmaceutical use: Available under the name Actimmune (Genentech). Used for reducing the frequency and severity of serious infections associated with chronic granulomatous disease (CGD).

Sequence similarities: Belongs to the type II (or gamma) interferon family.

Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IFN- gamma, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
