The Exonuclease I (ExoI) degrades single-stranded DNA in a 3'=>5' direction, releasing deoxyribonucleoside 5'-monophosphates in a stepwise manner and leaving 5'-terminal dinucleotides intact. It does not cleave DNA strands with terminal 3'-OH groups blocked by phosphoryl or acetyl groups (1).

Uniprot Description

Function: This enzyme is essential for phage DNA replication; it is believed to function in the removal of DNA-linked RNA primers. It is also necessary for host DNA degradation and phage genetic recombination.

Catalytic activity: Exonucleolytic cleavage in the 5'- to 3'-direction to yield nucleoside 5'-phosphates.
