Suitable for use in Western Blot.
Dilution: Western Blot: SCF migrates as a broad band between ~20 and 42kD in SDS-PAGE due to post-translational modifications. Unmodified SCF has a predicted molecular mass of 21.0kD. SCF separates into a number of isoforms with a pI between 3.0 and 7.0 in 2D PAGE due to post-translational modifications. This compares with the unmodified SCF with a predicted pI of 5.76.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Stem Cell Factor (SCF) is a hematopoietic growth factor that stimulates the proliferation of mast cells. SCF also mediates cell-cell adhesion and acts synergistically with other cytokines. The actions of SCF are exerted by binding to the SCF Receptor (PE-1243-50) which results in receptor dimerization. SCF is a type I membrane glycoprotein with 5 potential N-linked glycosylation sites. Soluble SCF is produced by proteolytic cleavage of the extracellular domain.

This gene encodes the ligand of the tyrosine-kinase receptor encoded by the KIT locus. This ligand is a pleiotropic factor that acts in utero in germ cell and neural cell development, and hematopoiesis, all believed to reflect a role in cell migration. In adults, it functions pleiotropically, while mostly noted for its continued requirement in hematopoiesis. Two transcript variants encoding different isoforms have been found for this gene. SCF: Ligand for the receptor-type protein-tyrosine kinase KIT. Plays an essential role in the regulation of cell survival and proliferation, hematopoiesis, stem cell maintenance, gametogenesis, mast cell development, migration and function, and in melanogenesis. KITLG/SCF binding can activate several signaling pathways. Promotes phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase, and subsequent activation of the kinase AKT1. KITLG/SCF and KIT also transmit signals via GRB2 and activation of RAS, RAF1 and the MAP kinases MAPK1/ERK2 and/or MAPK3/ERK1. KITLG/SCF and KIT promote activation of STAT family members STAT1, STAT3 and STAT5. KITLG/SCF and KIT promote activation of PLCG1, leading to the production of the cellular signaling molecules diacylglycerol and inositol 1,4,5- trisphosphate. KITLG/SCF acts synergistically with other cytokines, probably interleukins. Homodimer, non-covalently linked (Probable). Heterotetramer with KIT, binding two KIT molecules; thereby mediates KIT dimerization and subsequent activation by autophosphorylation. Belongs to the SCF family. 3 isoforms of the human protein are produced by alternative splicing.

It is sometimes possible for the material contained within the vial of "SCF, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening.
