Related Product Information for anti-HNF4A antibody
The protein encoded by this gene is a nuclear transcription factor which binds DNA as a homodimer. The encoded protein controls the expression of several genes, including hepatocyte nuclear factor 1 alpha, a transcription factor which regulates the expression of several hepatic genes. This gene may play a role in development of the liver, kidney, and intestines. Mutations in this gene have been associated with monogenic autosomal dominant non-insulin-dependent diabetes mellitus type I. Alternative splicing of this gene results in multiple transcript variants.

NCBI Description

The protein encoded by this gene is a nuclear transcription factor which binds DNA as a homodimer. The encoded protein controls the expression of several genes, including hepatocyte nuclear factor 1 alpha, a transcription factor which regulates the expression of several hepatic genes. This gene may play a role in development of the liver, kidney, and intestines. Mutations in this gene have been associated with monogenic autosomal dominant non-insulin-dependent diabetes mellitus type I. Alternative splicing of this gene results in multiple transcript variants encoding several different isoforms. [provided by RefSeq, Apr 2012]

Uniprot Description

Function: Transcriptionally controlled transcription factor. Binds to DNA sites required for the transcription of alpha 1-antitrypsin, apolipoprotein CIII, transthyretin genes and HNF1-alpha. May be essential for development of the liver, kidney and intestine.

Subunit structure: Homodimerization is required for HNF4-alpha to bind to its recognition site.

Subcellular location: Nucleus.

Post-translational modification: Phosphorylated on tyrosine residue(s); phosphorylation is important for its DNA-binding activity. Phosphorylation may directly or indirectly play a regulatory role in the subnuclear distribution. Phosphorylation at Ser-313 by AMPK reduces the ability to form homodimers and bind DNA. Ref.10 Ref.11 Ref.12Acetylation at Lys-458 lowers transcriptional activation by about two-fold.

Involvement in disease: Maturity-onset diabetes of the young 1 (MODY1) [MIM:125850]: A form of diabetes that is characterized by an autosomal dominant mode of inheritance, onset in childhood or early adulthood (usually before 25 years of age), a primary defect in insulin secretion and frequent insulin-independence at the beginning of the disease.Note: The disease is caused by mutations affecting the gene represented in this entry. Ref.14 Ref.15 Ref.17

Miscellaneous: Binds fatty acids.

Sequence similarities: Belongs to the nuclear hormone receptor family. NR2 subfamily.Contains 1 nuclear receptor DNA-binding domain.

Sequence caution: The sequence CAA54248.1 differs from that shown. Reason: Erroneous initiation. The sequence CAA61133.1 differs from that shown. Reason: Frameshift at positions 2 and 7. The sequence CAA61134.1 differs from that shown. Reason: Frameshift at positions 2 and 7. The sequence CAA61135.1 differs from that shown. Reason: Frameshift at positions 2 and 7.

Product Notes

The HNF4A hnf4a (Catalog #AAA649672) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The HNF4A, NT (Hepatocyte Nuclear Factor 4-alpha, HNF-4-alpha, Nuclear Receptor Subfamily 2 Group A Member 1, Transcription Factor 14, TCF-14, Transcription Factor HNF-4, HNF4, NR2A1, TCF14) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's HNF4A can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC) - Other applications not tested. Recommended Dilution: ELISA: 1:32,000 Western Blot: 0.1-0.5ug/ml Immunohistochemistry (FFPE): 2ug/ml Requires HIER citrate buffer, pH 6.0 Optimal dilutions to be determined by the researcher. Researchers should empirically determine the suitability of the HNF4A hnf4a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "HNF4A, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
