Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Do not freeze APC conjugates. Light sensitive. Aliquots are stable for at least 6 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

(Dictyostelium discoideum FAM21 contains a Capping Protein the interaction site (CPI) at amino acids 1201---1217. A construct was created where this site was removed from FAM21, and both the full length and FAM21ΔCPI were tagged with GFP and expressed in D.discoideum cells. GFP---trap beads were used to purify both constructs and western blotting was used to detect the presence of CAP32 (D. discoideum Capping Protein subunit). A GFP only construct was used as a control.It is shown here that CAP32 is pulled down with the full length protein but not when the interaction site is removed.)

NCBI Description

This gene encodes the beta subunit of the barbed-end actin binding protein, which belongs to the F-actin capping protein family. The capping protein is a heterodimeric actin capping protein that blocks actin filament assembly and disassembly at the fast growing (barbed) filament ends and functions in regulating actin filament dynamics as well as in stabilizing actin filament lengths in muscle and nonmuscle cells. A pseudogene of this gene is located on the long arm of chromosome 2. Multiple alternatively spliced transcript variants encoding different isoforms have been found.[provided by RefSeq, Aug 2013]

Uniprot Description

F-actin-capping proteins bind in a Ca2+-independent manner to the fast growing ends of actin filaments (barbed end) thereby blocking the exchange of subunits at these ends. Unlike other capping proteins (such as gelsolin and severin), these proteins do not sever actin filaments. Plays a role in the regulation of cell morphology and cytoskeletal organization.
