Western Blot (WB)

(Western blot shows lysates of A431 human epithelial carcinoma cell line, Jurkat human acute T cell leukemia cell line, and Daudi human Burkitt's lymphoma cell line untreated (-) or treated (+) with 1mM Pervanadate (PV) for 30 minutes. PVDF membrane was probed with 0.1ug/ml of Mouse Human Phospho-Paxillin (Y31) Antigen Affinity-purified Monoclonal Antibody followed by HRP-conjugated Mouse IgG Secondary Antibody. Specific bands were detected for Phospho-Paxillin (Y31) at approximately 65 to 68kD. This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.)

Testing Data

(Paxillin in HUVEC Human Cell Line. Paxillin phosphorylated at Y31 was detected in immersion fixed HUVEC human umbilical vein endothelial cells using Mouse Human Phospho-Paxillin (Y31) Antigen Affinity-purified Monoclonal Antibody at 10ug/ml for 3h at room temperature. Cells were stained using the NorthernLightsTM 557-conjugated Mouse IgG Secondary Antibody. Paxillin is a 65kD cytoskeletal adaptor protein and member of the Paxillin family. Human Paxillin is 591aa in length and contains four LIM zinc-binding domains. Alternative splicing produces three isoforms. Human Paxillin shares 94% and 85% aa identity with mouse and rat Paxillin, respectively. Paxillin is found at the interface between actin filaments and the plasma membrane, and it localizes to focal adhesions, where it provides a platform for the integration and coordination of adhesion-and growth factor-related signals. Paxillin phosphorylation at tyrosines 31 and 118 is required for integrin-mediated cytoskeletal reorganization, and may play a role in the disassembly of focal adhesions and stress fibers during cellular transformation.

Uniprot Description

PXN: a multi-domain cytoskeletal protein involved in actin-membrane attachment that localizes primarily to focal adhesion sites to the extracellular matrix. Phosphorylated by focal adhesion kinase (FAK) and is a component of integrin signaling. Its phosphorylation provides docking sites for recruitment of signaling molecules to focal adhesions. Binds in vitro to vinculin as well as to the SH3 domain of c-SRC and, when tyrosine phosphorylated, to the Crk SH2 domain. Interacts with GIT1, NUDT16L1/SDOS, PARVA and TGFB1I1. Component of cytoplasmic complexes, which also contain GIT1, ARHGEF6 and PAK1. Binds DDEF2. Interacts with unphosphorylated ITGA4. Interacts with RNF5.Three alternatively-spliced isoforms have been described. Isoform beta binds to focal adhesion kinase but weakly to vinculin. Isoform gamma binds to vinculin but only weakly to focal adhesion kinase.
