Solubility
Soluble in acidic buffer (maximum 4mg/ml), but difficult to dissolve in neutral buffer. In order to dissolve this product in neutral buffer, first dissolve in 0.01 or 0.05M acetic acid, and then add into neutral buffer containing 0.2M NaCl or dialyze against the same buffer.

Note
To avoid fibril formation under neutral conditions, it is recommended to use a buffer containing 0.15-0.2M sodium chloride and keep on ice. Type II collagen does not form a stable gel with incubation at 37°C. To prepare a type II collagen gel, addition components such as Type I collagen or other substances may be required.
Preparation and Storage
Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile buffer. Aliquot and store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Sensitive to light.

Collagen is an inert, rigid protein found predominantly in skin, ligaments, bones and teeth. Its most distinctive attribute, essential to a transmitter of mechanical force, is inelasticity. Its fundamental structural unit is tropo-collagen, a molecular rod about 2600Å in length and 15Å in diameter and 300,000 molecular weight. In tendons these macromolecules, grouped as collagen fibrils, run parallel to the axis. In skin the fibrils are interlaced and branched.

Highly purified type II collagen. Suitable for use as a standard for collagen analysis by SDS gel; as a substrate for MMP-1, MMP-8 and MMP-13; and as an antigen for immunizing animals for the induction of collagen-induced arthritis (CIA) in certain species and strains of experimental animals.

NCBI Description

This gene encodes the alpha-1 chain of type II collagen, a fibrillar collagen found in cartilage and the vitreous humor of the eye. Mutations in this gene are associated with achondrogenesis, chondrodysplasia, early onset familial osteoarthritis, SED congenita, Langer-Saldino achondrogenesis, Kniest dysplasia, Stickler syndrome type I, and spondyloepimetaphyseal dysplasia Strudwick type. In addition, defects in processing chondrocalcin, a calcium binding protein that is the C-propeptide of this collagen molecule, are also associated with chondrodysplasia. There are two transcripts identified for this gene. [provided by RefSeq, Jul 2008]

COL2A1: the alpha-1 chain of type II collagen, an extra-cellular matrix protein found in cartilage and the vitreous humor of the eye. It is essential for the normal embryonic development of the skeleton, for linear growth and for the ability of cartilage to resist compressive forces. Chondrocalcin is the calcium binding C-propeptide of this collagen molecule. Defects in this protein are associated with achondrogenesis, chondrodysplasia, early onset familial osteoarthritis, SED congenita, Langer-Saldino achondrogenesis, Kniest dysplasia, Stickler syndrome type I, and spondyloepimetaphyseal dysplasia Strudwick type. There are two transcripts identified for this gene. Belongs to the fibrillar collagen family. 3 isoforms of the human protein are produced by alternative splicing.
