Application Notes
Suitable for use in Direct ELISA, Western Blot and Flow Cytometry.
Dilution: Direct ELISA: 0.5-1.0ug/ml. The detection limit is ~1ng/well.
Western Blot: 0.1-0.2ug/ml. The detection limit is ~1ng/lane under non-reducing and reducing conditions.
Flow Cytometry: 50ug/ml. 10ul labels 1-2.5x10e5 cells in a total reaction volume not exceeding 200ul. Tested on rat splenocytes.

Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

HLA-G belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta-2 microglobulin). The heavy chain is anchored in the membrane. HLA-G is expressed on fetal derived placental cells. The heavy chain is approximately 45 kDa and its gene contains 8 exons. Exon one encodes the leader peptide, exons 2 and 3 encode the alpha1 and alpha2 domain, which both bind the peptide, exon 4 encodes the alpha3 domain, exon 5 encodes the transmembrane region, and exon 6 encodes the cytoplasmic tail.
