Form/Format
A sterile filtered antibody solution was lyophilized from PBS, pH 7.2
Applicable Applications for anti-CTGF antibody
Immunohistochemistry (IHC), Western Blot (WB), Neutrilization, Sandwich ELISA
Application Notes
Immunohistochemistry:
This antibody stained formalin-fixed, paraffin-embedded sections of human breast invasive malignant carcinoma. The recommended concentration is 0.125 ug/mL-0.250 ug/mL with an overnight incubation at 4°C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 sodium citrate buffer is recommended. Optimal concentrations and conditions may vary.
Neutrilization:
To yield one-half maximal inhibition [ND50] of the biological activity of human CTGF (5.0 ug/ml), a concentration of 6.0 - 9.0 ug/ml of this antibody is required.
Sandwich ELISA:
To detect Human CTGF by sandwich ELISA (using 100 ul/well antibody solution) a concentration of 0.5 - 2.0 ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human CTGF (MBS695113) as a detection antibody, allows the detection of at least 0.2 - 0.4 ng/well of recombinant Human CTGF.
Western Blot:
To detect Human CTGF by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hCTGF is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions. Optimal dilutions should be determined by each laboratory for each application.
Preparation
Produced from sera of rabbits immunized with highly pure recombinant Human CTGF. Anti-human CTGF specific antibody was purified by affinity chromatography employing an immobilized Human CTGF matrix.
Immunogen
E.Coli derived recombinant Human CTGF (Please Inquire).
Reconstitution
Centrifuge vial prior to opening. Reconstitute in sterile water to a concentration of 0.1-1.0mg/ml.
Preparation and Storage
Store at -20°C.
Related Product Information for anti-CTGF antibody
CTGF is a 38kD, cysteine-rich, secreted peptide. It is a new member of the peptide family that include serum-induced immediate early gene products, a v-src-induced peptide, and a putative proto-oncogene. Among the many functions of the CTGF gene family are embryogenesis, wound healing, and regulation of extracellular matrix production.

NCBI Description

The protein encoded by this gene is a mitogen that is secreted by vascular endothelial cells. The encoded protein plays a role in chondrocyte proliferation and differentiation, cell adhesion in many cell types, and is related to platelet-derived growth factor. Certain polymorphisms in this gene have been linked with a higher incidence of systemic sclerosis. [provided by RefSeq]

Uniprot Description

CTGF: Major connective tissue mitoattractant secreted by vascular endothelial cells. Promotes proliferation and differentiation of chondrocytes. Mediates heparin- and divalent cation-dependent cell adhesion in many cell types including fibroblasts, myofibroblasts, endothelial and epithelial cells. Enhances fibroblast growth factor-induced DNA synthesis. Belongs to the CCN family. 2 isoforms of the human protein are produced by alternative splicing.

Optimal dilutions should be determined by each laboratory for each application. Researchers should empirically determine the suitability of the CTGF ctgf for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "CTGF, Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
