Purified by Protein G affinity chromatography.
Dilution: Immunohistochemistry: Reported to stain SAF-B in paraf n tissue sections.
Immunocytochemistry: Reported to stain SAF-B in MCF-7 cells.
Immunoprecipitation: Reported to immunoprecipitate SAF-B from MCF-7 and HepG2 cell lysates.
Western Blot: 0.5ug/ml detects SAF-B in whole cell extracts or RIPA lysates from MCF-7 cells. MCF-7 whole cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S0430 (0.5ug/ml). Proteins were visualized using an IgG (HRP) goat anti-mouse* and a chemiluminescence detection system.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a DNA-binding protein that has high specificity for scaffold or matrix attachment region DNA elements (S/MAR DNA). This protein is thought to be involved in attaching the base of chromatin loops to the nuclear matrix but there is conflicting evidence as to whether this protein is a component of chromatin or a nuclear matrix protein. Scaffold attachment factors are a specific subset of nuclear matrix proteins (NMP) that specifically bind to S/MAR. This encoded protein is thought to serve as a molecular base to assemble a 'transcriptosome complex' in the vicinity of actively transcribed genes. It is involved in the regulation of the heat shock protein 27 transcription and also can act as an estrogen receptor corepressor. This gene is a candidate gene for breast tumorigenesis. Uniprot Description
SAFB1: Binds to scaffold/matrix attachment region (S/MAR) DNA and forms a molecular assembly point to allow the formation of a 'transcriptosomal' complex (consisting of SR proteins and RNA polymerase II) coupling transcription and RNA processing. Can function as an estrogen receptor corepressor and can also bind to the HSP27 promoter and decrease its transcription. When associated with RBMX, binds to and stimulates transcription from the SREBF1 promoter. Can inhibit cell proliferation.
Protein type: RNA-binding; Nuclear receptor co-regulator; DNA-binding
Chromosomal Location of Human Ortholog: 19p13.3-p13.2
Cellular Component: nucleoplasm; nucleus
Molecular Function: protein binding; double-stranded DNA binding; nucleotide binding; chromatin binding
Biological Process: establishment and/or maintenance of chromatin architecture; estrogen receptor signaling pathway; transcription, DNA-dependent; hormone metabolic process; positive regulation of transcription from RNA polymerase II promoter; growth

Researchers should empirically determine the suitability of the SAFB safb for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Scaffold Attachment Factor B, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
