Application Notes
Suitable for use in ELISA, Immunohistochemistry, Immunocytochemistry, Immunoprecipitation and Western Blot.
Dilution: Immunohistochemistry: Reported to stain SAF-B in paraf n tissue sections.
Immunocytochemistry: Reported to stain SAF-B in MCF-7 cells.
Immunoprecipitation: Reported to immunoprecipitate SAF-B from MCF-7 and HepG2 cell lysates.
Western Blot: 0.5ug/ml detects SAF-B in whole cell extracts or RIPA lysates from MCF-7 cells. MCF-7 whole cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S0430 (0.5ug/ml). Proteins were visualized using an IgG (HRP) goat anti-mouse* and a chemiluminescence detection system.
Immunogen
Synthetic peptide corresponding to aa345-357 (CPEARDKEDGRKF) from the human Scaffold Attachment Factor B conjugated to KLH (U72355). Sequence Homology: Mouse and human T-box protein 2, (8/11 identical aa).
Preparation and Storage
May be stored at 4 degree C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

SAFB1: Binds to scaffold/matrix attachment region (S/MAR) DNA and forms a molecular assembly point to allow the formation of a 'transcriptosomal' complex (consisting of SR proteins and RNA polymerase II) coupling transcription and RNA processing. Can function as an estrogen receptor corepressor and can also bind to the HSP27 promoter and decrease its transcription. When associated with RBMX, binds to and stimulates transcription from the SREBF1 promoter. Can inhibit cell proliferation.
