Application Notes
Western Blot: This antibody can be used at 1-2ug/ml with the appropriate secondary reagents to detect human CRHBP. Using a colorimetric detection system, the detection limit for rhCRHBP is approximately 5 ng/lane under non-reducing and reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold., Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect human CRHBP. The detection limit for rhCRHBP is approximately 15 ng/well., Optimal dilutions should be determined by each laboratory for each application.

Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. Reconstitute to nominal volume by adding sterile PBS with 40-50% glycerol and store at -20 degree C. Reconstituted product is stable for 12 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Human CRHBP is a 37kD secreted glycoprotein that likely serves to regulate CRH availability to the adrenal. In human, CRHBP complexes with CRH to form a circulating 41kD complex. In rodent and horse, this complex is restricted to the CNS. Human CRHBP is synthesized as a 322 amino acid (aa) precursor that contains a 24 aa signal sequence and a 298 aa mature region. The mature region contains one potential CUB domain and five intrachain disulfide bonds. Human mature CRHBP shares 87% and 84% sequence identity with mouse and equine CRHBP, respectively.

NCBI Description

Corticotropin-releasing hormone is a potent stimulator of synthesis and secretion of preopiomelanocortin-derived peptides. Although CRH concentrations in the human peripheral circulation are normally low, they increase throughout pregnancy and fall rapidly after parturition. Maternal plasma CRH probably originates from the placenta. Human plasma contains a CRH-binding protein which inactivates CRH and which may prevent inappropriate pituitary-adrenal stimulation in pregnancy. [provided by RefSeq]
