Liquid. In 25mM NaBorate, pH9.3, containing 5mM CaCl2, 20uM ZnCl2, 0.05% Brij-35 and 20% glycerol
Source Note
Active recombinant MMP-19. Recombinant MMP-19 corresponding to aa Leu99-Ser259 (NM_002429), which comprises the catalytic domain of human MMP-19, with a C-terminal purification tag
Activity
Preincubation of MMP-19 catalytic domain at 1.9uM with the broad-spectrum inhibitor GM6001 at 100nM for 30 minutes only weakly inhibits enzymatic activity (27%)
Specific Activity
>= 0.40U/ug. One U=100 pmol/min at 37 degree C using the colorimetric thiopeptolide Ac-Pro-Leu-Gly-S-Leu-Leu-Gly-OEt (100uM) as substrate
Dry Ice Shipment
Extra charge fee may add to your shipping cost as dry ice is required to ship this product.
Preparation and Storage
For long term storage, store at -80 degree C
When stored under the recommended conditions, this enzyme is stable at the concentration supplied, in its current storage buffer. Procedures such as dilution followed by refreezing could lead to loss of activity
Avoid freeze/thaw cycles

Uniprot Description

MMP19: Endopeptidase that degrades various components of the extracellular matrix, such as aggrecan and cartilage oligomeric matrix protein (comp), during development, haemostasis and pathological conditions (arthritic disease). May also play a role in neovascularization or angiogenesis. Hydrolyzes collagen type IV, laminin, nidogen, nascin-C isoform, fibronectin, and type I gelatin. May play a role in pathological processes participating in rheumatoid arthritis (RA)-associated joint tissue destruction. Autoantigen anti-MMP19 are frequent in RA patients. Belongs to the peptidase M10A family. 3 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted; EC 3.4.24.-; Secreted, signal peptide; Protease

Chromosomal Location of Human Ortholog: 12q14

Cellular Component: proteinaceous extracellular matrix; extracellular region

Molecular Function: zinc ion binding; metalloendopeptidase activity; calcium ion binding

Biological Process: collagen catabolic process; extracellular matrix disassembly; extracellular matrix organization and biogenesis; response to cAMP; luteolysis; ovarian follicle development; response to hormone stimulus; angiogenesis; ovulation from ovarian follicle; cell differentiation; proteolysis

Product Notes

The MMP-19 mmp19 (Catalog #AAA566700) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "MMP-19, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
