Biological Significance
PKC zeta is an atypical isoform of the PKC family. PKC zeta is found in both particulate and soluble fractions and cannot be activated by phorbol ester. Treatment of cells with phorbol ester which activates PKCalpha, gamma, delta, and e isoforms in NIH3T3 cells significantly reduced proliferation of cells. Overexpression of PKC zeta and subsequent phorbol ester treatment abolished phorbol ester- induced reduction in cell proliferation. Overexpression of PKC zeta also potentiated phorbol ester- induced mitogen-activated protein kinase activation in a PKC-dependent manner. The effects of PKC zeta overexpression on proliferation and MAP kinase activation are proportional to the levels of PKC zeta expression. PKC zeta as an upstream modulator of p70S6K, an important regulator of cell proliferation.
