Specificity
Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

NCBI Description

The protein encoded by this gene is a member of the chromogranin/secretogranin family of neuroendocrine secretory proteins. It is found in secretory vesicles of neurons and endocrine cells. This gene product is a precursor to three biologically active peptides; vasostatin, pancreastatin, and parastatin. These peptides act as autocrine or paracrine negative modulators of the neuroendocrine system. Two other peptides, catestatin and chromofungin, have antimicrobial activity and antifungal activity, respectively. Two transcript variants encoding different isoforms have been found for this gene. Uniprot Description

CHGA: a member of the chromogranin/secretogranin family of neuroendocrine secretory proteins. It is found in secretory vesicles of neurons and endocrine cells and is a precursor to multiple biologically active peptides including vasostatin, pancreastatin, and chromostatin. Binds calcium with a low-affinity. Vasostatin negatively regulates angiogenesis and has antibacterial activity against some Gram-positive bacteria. Pancreastatin inhibits glucose induced insulin release from the pancreas. Chromostatin inhibits catecholamine release from chromaffin cells.

Protein type: Vesicle; Secreted; Cell adhesion; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 14q32

Cellular Component: extracellular space; perinuclear region of cytoplasm; secretory granule

Biological Process: defense response to Gram-negative bacterium; defense response to Gram-positive bacterium; mast cell activation; mast cell chemotaxis; mast cell cytokine production; mast cell degranulation; negative regulation of catecholamine secretion; negative regulation of vasodilation; organelle organization and biogenesis; positive regulation of cAMP metabolic process; regulation of blood pressure; regulation of the force of heart contraction

Product Notes

The CHGA chga (Catalog #AAA438119) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The Chromogranin A / CHGA (Neuroendocrine Marker) Mouse Monoclonal Antibody reacts with Human, Monkey. Does not react with Mouse and Rat and may cross-react with other species as described in the data sheet. AAA Biotech's Chromogranin A / CHGA can be used in a range of immunoassay formats including, but not limited to, Flow Cytometry (FC/FACS), Immunofluorescence (IF), Immunohistochemistry (IHC) Formalin. Flow Cytometry (0.5-1ug/million cells in 0.1ml) Immunofluorescence (0.5-1ug/ml) Immunohistochemistry (Formalin-fixed) (0.5-1ug/ml for 30 minutes at RT) (Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes). Researchers should empirically determine the suitability of the CHGA chga for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Chromogranin A / CHGA, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
