Pre-Treatment
After de-waxing the tissue slices they are treated with 0.2% hyaluronidase (app. 300 U/mg) in TBS 15 min at 37 degree C. There after non-specific binding is blocked by blocking serum or 3% BSA in TBS. For peroxidase systems blocking with 1% peroxide solution in TBS for 30 min at RT is recommended.

Related Product Information for anti-Fn1 antibody
This antibody may be used to identify fibronectin during wound healing, during tumour progression, and tumour invasion respectively. Fibronectins are glycoproteins, which occur as dimers of two 200 kDa subunits. They have binding domains for bacterial proteins, collagens, heparin-like molecules and fibrin. Cellular fibronectin is widely distributed in the stroma of malignant tumours. Murine fibronectin 100%, murine collagen type I, III and IV <0.1%, murine laminin <0.1% (RIA at 1:10000 dilution).

Uniprot Description

Fibronectins bind cell surfaces and various compounds including collagen, fibrin, heparin, DNA, and actin. Fibronectins are involved in cell adhesion, cell motility, opsonization, wound healing, and maintenance of cell shape healing, and maintenance of cell shape. Involved in osteoblast compaction through the fibronectin fibrillogenesis cell-mediated matrix assembly process, essential for osteoblast mineralization. Participates in the regulation of type I collagen deposition by osteoblasts.

Product Notes

The Fn1 fn1 (Catalog #AAA397211) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Fibronectin, mouse reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's Fibronectin can be used in a range of immunoassay formats including, but not limited to, Immunofluorescence (IF), Immunohistochemistry (IHC) Paraffin, ELISA (EIA), Radioimmunoassay (RIA). IFA: 1:80 IHC-P: 1:1000 ELISA: 1:200. Researchers should empirically determine the suitability of the Fn1 fn1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Fibronectin, Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
