SDS-Page (IkBa mAb tested by Western blot. The anti-IkBa detects both phosphorylated and non-phosphorylated forms of IkBa by Western blot. Jurkat cells (~1 x 107) were treated for indicated time periods with 10 nm/ml PMA, 1 uM ionomycin and anti-CD28 (1:10,000 dilution). 10 ul of cytoplasmic (C) and nuclear extract (N) were resolved on a 8.75% SDS-PAGE and transferred to Immobilon membrane. Related Product Information for anti-IkBa antibody
Short Description: Our IkBa antibody (mAb)(Clone 6A920) was raised in a Mouse host. It has been validated for use in Western blot; it has been shown to react with Human and Mouse samples.

Background: IkBa-NFkB (NFkB p50 & NFkB p65) signaling is controlled to a large extent by the sequestration of the NFkB complex in the cytoplasm by its association with one of the IkB family of proteins. IkBa is phosphorylated at Ser32 and Ser36 (IkBa phospho Ser32,36) by the IkB Kinase (IKK) complex, resulting in the degradation of IkB and the nuclear translocation of NFkB.

NCBI Description

This gene encodes a member of the NF-kappa-B inhibitor family, which contain multiple ankrin repeat domains. The encoded protein interacts with REL dimers to inhibit NF-kappa-B/REL complexes which are involved in inflammatory responses. The encoded protein moves between the cytoplasm and the nucleus via a nuclear localization signal and CRM1-mediated nuclear export. Mutations in this gene have been found in ectodermal dysplasia anhidrotic with T-cell immunodeficiency autosomal dominant disease. [provided by RefSeq, Aug 2011]

Uniprot Description

IkB-alpha: a regulatory protein that inhibits NF-kappa-B by complexing with and trapping it in the cytoplasm. May be involved in regulation of transcriptional responses to NF-kappa-B, including cell adhesion, immune and proinflammatory responses, apoptosis, differentiation and growth. Controlled by sequential serine-phosphorylation, ubiquitination and degradation.

Product Notes

The IkBa nfkbia (Catalog #AAA388667) is an Antibody produced from Mouse and is intended for research purposes only. The product is available for immediate purchase. The IkBa antibody (mAb)(Clone 6A920) reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's IkBa can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Applications Validated: WB: 1-2ug/ml dilution For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all blocking solutions may also reduce background. Researchers should empirically determine the suitability of the IkBa nfkbia for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "IkBa, Monoclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
