Sensitivity

The sensitivity, minimum detectable dose of IL-31 using this IL-31 ELISA kit was found to be <18.3pg/ml. This was determined by adding 2 standard deviations to the mean OD obtained when the zero standard was assayed 35 times (6 independent assays)

Intra-assay Precision

Reproducibility within the assay was evaluated in three independent experiments. Each assay was carried out with 6 replicates (3 duplicates) of 6 different samples containing different concentrations of IL-31. Data below show the mean concentration and the coefficient of variation for each sample. The overall intraassay coefficient of variation has been calculated to be 2.5%.

Inter-assay Precision

Assay to assay reproducibility within one laboratory will be evaluated in three independent experiments by two technicians. Each assay will be carried out with 6 replicates (3 duplicates) in 2 human pooled serum, 2 in RPMI and 2 in standard diluent with samples containing different concentrations of IL-31. 2 standard curves were run on each plate. The calculated overall coefficient of variation was 5.2%.

IL31, which is made principally by activated Th2-type T cells, interacts with a heterodimeric receptor consisting of IL31RA (MIM 609510) and OSMR (MIM 601743) that is constitutively expressed on epithelial cells and keratinocytes. IL31 may be involved in the promotion of allergic skin disorders and in regulating other allergic diseases, such as asthma (Dillon et al., 2004 [PubMed 15184896]).[supplied by OMIM, Mar 2008]
