Small guanosine triphosphatases, typified by the mammalian Ras proteins, play major roles in the regulation of numerous cellular pathways. IkappaBinteracting Ras-like proteins, kappaB-Ras1 and kappaB-Ras2, belong to a subclass of evolutionarily conserved Ras-like proteins that differ from other Ras proteins in containing amino acids at positions 12 and 61 that are similar to those present in the oncogenic forms of Ras. These Ras-like proteins, kappaB-Ras1 and kappaB-Ras2, interact with the PEST domains of IkappaBa and IkappaBb and decreasetheir rate of degradation. kappaB-Ras2 shows 71% identity to kappaB-Ras1. In cells,kappaB-Ras proteins are associated only with NFkappaB:IkappaBb complexes and therefore may provide an explanation for the slower rate of degradation of IkappaBb compared with IkappaBa.

NKIRAS1: Atypical Ras-like protein that acts as a potent regulator of NF-kappa-B activity by preventing the degradation of NF-kappa-B inhibitor beta (NFKBIB) by most signals, explaining why NFKBIB is more resistant to degradation. May act by blocking phosphorylation of NFKBIB and mediating cytoplasmic retention of p65/RELA NF-kappa-B subunit. It is unclear whether it acts as a GTPase. Both GTP- and GDP-bound forms block phosphorylation of NFKBIB. Belongs to the small GTPase superfamily. Ras family. KappaB-Ras subfamily.
