Immunofluorescence (IF)

(IF image of MBS3001157 stained A375 cells. The cells were 4% paraformaldehyde fixed (20 min) and then incubated in 10% normal goat serum for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody Tubulin beta (4D9) mAb # MBS3001157 (1:200) at 10ug/ml overnight at +4 degree C. The secondary antibody (Green) was Goat Anti-Mouse IgG (H+L) FITC used at a 1/400 dilution for 1h. Hoechst33342 was used to stain the cell nuclei (blue).)

Microtubules are constituent parts of the mitotic apparatus, cilia, flagella, and elements of the cytoskeleton. They consist principally of 2 soluble proteins, alpha- and beta-tubulin, each of about 55, 000 kDa. Antibodies against beta Tubulin are useful as loading controls for Western Blotting. However it should be noted that levels of beta Tubulin may not be stable in certain cells.

NCBI Description

This gene encodes a class III member of the beta tubulin protein family. Beta tubulins are one of two core protein families (alpha and beta tubulins) that heterodimerize and assemble to form microtubules. This protein is primarily expressed in neurons and may be involved in neurogenesis and axon guidance and maintenance. Mutations in this gene are the cause of congenital fibrosis of the extraocular muscles type 3. Alternate splicing results in multiple transcript variants. A pseudogene of this gene is found on chromosome 6. [provided by RefSeq, Oct 2010]

TUBB3: Tubulin is the major constituent of microtubules. It binds two moles of GTP, one at an exchangeable site on the beta chain and one at a non-exchangeable site on the alpha-chain. TUBB3 plays a critical role in proper axon guidance and mantainance. Dimer of alpha and beta chains. Expression is primarily restricted to central and peripheral nervous system. Greatly increased expression in most cancerous tissues. Belongs to the tubulin family.
