Description: This assay employs a two-site sandwich ELISA to quantitate FAM90A9 in samples. An antibody specific for FAM90A9 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FAM90A9 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FAM90A9 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FAM90A9 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: FAM90A9 belongs to subfamily II of the primate-specific FAM90A gene family, which originated from multiple duplications and rearrangements. For background information on the FAM90A gene family, as well as information on the evolution of FAM90A genes, The deduced 434-amino acid FAM90A9 protein contains a CCHC-type zinc finger motif, suggesting that it may interact with RNA or DNA. RT-PCR using primers that did not distinguish between individual FAM90A genes suggested that members of FAM90A subfamily II, such as FAM90A9, are ubiquitously expressed. FAM90A9 gene contains 5 exons. Exon 1 is noncoding and contains a LINE element, L1MB3, and the sequence 3-prime of the last exon includes a long terminal repeat of the ERV-K family. Like other members of FAM90A subfamily II, FAM90A9 lacks repetitive sequences at its 5-prime end.
