Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate IL12RB2 in samples. An antibody specific for IL12RB2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyIL12RB2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL12RB2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL12RB2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

The protein encoded by this gene is a type I transmembrane protein identified as a subunit of the interleukin 12 receptor complex. The coexpression of this and IL12RB1 proteins was shown to lead to the formation of high-affinity IL12 binding sites and reconstitution of IL12 dependent signaling. The expression of this gene is up-regulated by interferon gamma in Th1 cells, and plays a role in Th1 cell differentiation. The up-regulation of this gene is found to be associated with a number of infectious diseases, such as Crohn's disease and leprosy, which is thought to contribute to the inflammatory response and host defense. Several transcript variants encoding different isoforms and non-protein coding transcripts have been found for this gene. [provided by RefSeq, Apr 2012]

Uniprot Description

IL12RB2: Receptor for interleukin-12. This subunit is the signaling component coupling to the JAK2/STAT4 pathway. Promotes the proliferation of T-cells as well as NK cells. Induces the promotion of T-cells towards the Th1 phenotype by strongly enhancing IFN-gamma production. Belongs to the type I cytokine receptor family. Type 2 subfamily. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Cell cycle regulation; Membrane protein, integral; Receptor, cytokine

Chromosomal Location of Human Ortholog: 1p31.3

Cellular Component: external side of plasma membrane; integral component of plasma membrane; plasma membrane

Molecular Function: hematopoietin/interferon-class (D200-domain) cytokine receptor activity; protein kinase binding

Biological Process: cell surface receptor signaling pathway; interferon-gamma production; peptidyl-tyrosine phosphorylation; positive regulation of cell proliferation; positive regulation of interferon-gamma production; response to lipopolysaccharide

Product Notes

The Human IL12RB2 il12rb2 (Catalog #AAA285508) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA285508 ELISA Kit recognizes Human IL12RB2. It is sometimes possible for the material contained within the vial of "Interleukin-12, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
