Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CRHR1 in samples. An antibody specific for CRHR1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCRHR1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CRHR1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CRHR1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description
G-protein coupled receptor for CRH (corticotropin-releasing factor) and UCN (urocortin). Has high affinity for CRH and UCN. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and down-stream effectors, such as adenylate cyclase. Promotes the activation of adenylate cyclase, leading to increased intracellular cAMP levels. Inhibits the activity of the calcium channel CACNA1H. Required for normal embryonic development of the adrenal gland and for normal hormonal responses to stress. Plays a role in the response to anxiogenic stimuli.
