Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate CDKN1A in samples. An antibody specific for CDKN1A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyCDKN1A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CDKN1A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CDKN1A bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

May be involved in p53/TP53 mediated inhibition of cellular proliferation in response to DNA damage. Binds to and inhibits cyclin-dependent kinase activity, preventing phosphorylation of critical cyclin-dependent kinase substrates and blocking cell cycle progression. Functions in the nuclear localization and assembly of cyclin D-CDK4 complex and promotes its kinase activity towards RB1. At higher stoichiometric ratios, inhibits the kinase activity of the cyclin D-CDK4 complex. Inhibits DNA synthesis by DNA polymerase delta by competing with POLD3 for PCNA binding.
