ZNF426 (zinc finger protein 426), also known as MGC2663, is a 554 amino acid protein that is thought to be involved in transcriptional regulation. Localized to the nucleus, ZNF426 contains one KRAB domain and 12 C2H2-type zinc fingers through which it may convey DNA, RNA and protein binding capabilities. Specifically, ZNF426 may interact with the viral protein KSHV ORF 50 and, through this interaction, may activate viral gene transcription.
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZNF426 in samples. An antibody specific for ZNF426 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZNF426 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZNF426 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZNF426 bound in the initial step. The color development is stopped and the intensity of the color is measured.
