The 402-amino acid Zcchc12 protein contains an N-terminal paraneoplastic antigen (MA)-like region, a C2HC-type zinc finger motif, and a putative nuclear localization signal. Northern blot analysis detected restricted expression of Zcchc12 in the ventral forebrain (including septum, amygdala, putamen, thalamus, and hypothalamus) of E11.5-E12.5 mice, and predominant expression in the brain and lower expression in the testis of adult mice.
Zcchc12 protein binds directly to Smad1. Zcchc12 functions as a transcriptional coactivator necessary for bone morphogenetic protein (BMP)-dependent, cholinergic-neuron-specific gene expression.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZCCHC12 in samples. An antibody specific for ZCCHC12 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZCCHC12 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZCCHC12 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZCCHC12 bound in the initial step. The color development is stopped and the intensity of the color is measured.
