Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate KLKB1 in samples. An antibody specific for KLKB1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyKLKB1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KLKB1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KLKB1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Uniprot Description

KLKB1: The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin. Defects in KLKB1 are the cause of prekallikrein deficiency (PKK deficiency); also known as Fletcher factor deficiency. This disorder is a blood coagulation defect. Belongs to the peptidase S1 family. Plasma kallikrein subfamily.
