Intended Uses: For the quantitative detection of Human Potassium/sodium hyperpolarization -activated cyclic nucleotide-gated channel 1 (HCN1) concentration in serum, plasma and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate HCN1 in Human serum, plasma. An antibody specific for HCN1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HCN1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HCN1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HCN1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The membrane protein encoded by this gene is a hyperpolarization-activated cation channel that contributes to the native pacemaker currents in heart and neurons. The encoded protein can homodimerize or heterodimerize with other pore-forming subunits to form a potassium channel. This channel may act as a receptor for sour tastes.
