This assay employs a two-site sandwich ELISA to quantitate ARHGEF11 in samples. An antibody specific for ARHGEF11 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ARHGEF11 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ARHGEF11 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ARHGEF11 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Rho GTPases play a fundamental role in numerous cellular processes that are initiated by extracellular stimuli that work through G protein coupled receptors. The encoded protein may form a complex with G proteins and stimulate Rho-dependent signals. A similar protein in rat interacts with glutamate transporter EAAT4 and modulates its glutamate transport activity. Expression of the rat protein induces the reorganization of the actin cytoskeleton and its overexpression induces the formation of membrane ruffling and filopodia.
