Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Wingless Type MMTV Integration Site Family, Member 10B (WNT10B) were tested on 3 different plates, 8 replicates in each plate.
CV(%) = SD/meanX100
Intra-Assay: CV<10%
Inter-Assay: CV<12%
Test Principle
The microplate provided in this kit has been pre-coated with an antibody specific to Wingless Type MMTV Integration Site Family, Member 10B (WNT10B). Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to Wingless Type MMTV Integration Site Family, Member 10B (WNT10B). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then the mixture of substrate A and B is added to generate glow light emission kinetics. Upon plate development, the intensity of the emitted light is proportional to the Wingless Type MMTV Integration Site Family, Member 10B (WNT10B) level in the sample or standard.
Preparation and Storage
The stability of kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition.
To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.

NCBI Description

The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It may be involved in breast cancer, and its protein signaling is likely a molecular switch that governs adipogenesis. This protein is 96% identical to the mouse Wnt10b protein at the amino acid level. This gene is clustered with another family member, WNT1, in the chromosome 12q13 region. [provided by RefSeq, Jul 2008]

Uniprot Description

WNT10B: Ligand for members of the frizzled family of seven transmembrane receptors. Probable developmental protein. May be a signaling molecule which affects the development of discrete regions of tissues. Is likely to signal over only few cell diameters. Defects in WNT10B are the cause of split-hand/foot malformation type 6 (SHFM6). SHFM is a limb malformation involving the central rays of the autopod and presenting with syndactyly, median clefts of the hands and feet, and aplasia and/or hypoplasia of the phalanges, metacarpals, and metatarsals. SHFM6 is a autosomal recessive disorder. Belongs to the Wnt family.

Protein type: Secreted; Secreted, signal peptide

Product Notes

The Human WNT10B wnt10b (Catalog #AAA2709244) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA2709244 CLIA Kit recognizes Human WNT10B. AAA Biotech's Wingless Type MMTV Integration Site Family, Member 10B (WNT10B) can be used in a range of immunoassay formats including, but not limited to, Chemiluminescence Immunoassay (CLIA). Researchers should empirically determine the suitability of the WNT10B wnt10b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Wingless Type MMTV Integration Site Family, Member 10B (WNT10B), CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
