Specificity
This item is specific for glutamate receptor subunit 2 (GluR2), a component of the AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid) group of ionotropic glutamate receptors, which play a key role at excitatory synapses, including synaptic transmission, stabilisation and plasticity.

GluR2 is possibly the most important of the AMPA receptor subunits, responsible for AMPA receptor rectifying properties, control of ion flow and in particular the influx of calcium. The majority of GluR2 in the CNS is expressed in the GluR2(R) form, containing a critical arginine residue (as opposed to a glutamine residue) in the Transmembrane region 2 (TM2) domain, thereby rendering native AMPA receptors impermeable to calcium.

NCBI Description

Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. This gene product belongs to a family of glutamate receptors that are sensitive to alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA), and function as ligand-activated cation channels. These channels are assembled from 4 related subunits, Gria1-4. The subunit encoded by this gene (Gria2) is subject to RNA editing (Q/R and R/G), which is thought to render the channels impermeable to Ca(2+), and to affect the kinetic aspects of these channels in rat brain. Alternative splicing, resulting in transcript variants encoding different isoforms (flip and flop), has been noted for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

GluR2: an integral membrane protein belonging to the glutamate-gated ion channel family. L-glutamate (Glu) acts as an excitatory neurotransmitter at many synapses in the central nervous system. Glutamate receptors are heteromeric protein complexes with multiple subunits, each possessing transmembrane regions, and all arranged to form a ligand-gated ion channel. The postsynaptic actions of Glu are mediated by a variety of receptors that are named according to their selective agonists. This receptor binds AMPA(quisqualate) > glutamate > kainate. Each of the four GluR proteins (GRIA1-4) include flip and flop isoforms generated by alternative RNA splicing.

Product Notes

The GluR2 gria2 (Catalog #AAA221864) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI GluR2 reacts with Chicken (Expected from Sequence), Human, Monkey (Expected from Sequence), Mouse (Expected from Sequence), Zebrafish (Expected from Sequence) and may cross-react with other species as described in the data sheet. AAA Biotech's GluR2 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Western Blot: This item detects a band of approximately 100 kDa in rat brain hippocampus cell lysates. Western Blotting: Maximum Dilution: 1/1000. Researchers should empirically determine the suitability of the GluR2 gria2 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "GluR2, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
