Application Notes
ELISA: This purified human IL-10antibody may be used in an indirect ELISA or as the capture reagent in a sandwich ELISA with our biotinylated human IL-10 antibody as the detection reagent and recombinant human IL-10 as the standard.
Western Blot: This antibody may be used in Western Blotting under either reducing or non-reducing conditions with recombinant human IL-10 as the positive control.
ELISA: Maximum Dilution: 0.5ug/ml
Functional Assays: Minimum Dilution: 4.0; Maximum Dilution: 6.0ug/ml
Western Blotting: Minimum Dilution: 0.1; Maximum Dilution: 0.2ug/ml
Reconstitution
Reconstitute with 0.1ml distilled water Care should be taken during reconstitution as the protein may appear as a film at the bottom of the vial. MyBioSource recommends that the vial is gently mixed after reconstitution.For long term storage the addition of 0.09% sodium azide is recommended. N.B. For functional studies do not add sodium azide.

Preparation and Storage
Prior to reconstitution store at 4 degree C. After reconstitution store at -20 degree C. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 12 months from date of reconstitution.

Rabbit anti Human Interleukin-10 antibody recognizes human IL-10 (Interleukin-10), an 18.5kDa immunosuppressive cytokine originally known as cytokine synthesis inhibiting factor (CSIF), which specifically regulates immune-mediated inflammation and shares high sequence homology with murine IL-10 and the viral protein BCRF1. IL-10, produced by macrophages, mast cells, T cells, activated B cells and keratinocytes, inhibits the secretion of a variety of pro-inflammatory cytokines including TNFalpha, IL-1, IL-2, IL-3 and IL-8 and is chemotactic for CD8+ T cells. The suppressive ability of IL-10 has become a focus area for the treatment of Crohn's disease, but with conflicting results.

NCBI Description

The protein encoded by this gene is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immunoregulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immunoregulator in the intestinal tract. Mutations in this gene are associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis.[provided by RefSeq, May 2011]
