Goat
Reactivity
Dog (Expected from Sequence), Mouse (Expected from Sequence), Rat (Expected from Sequence)
Antibody reactivity and working conditions may vary between species.
Clonality
Polyclonal
Isotype
IgG
Specificity
This item recognises an epitope within the C-terminal (CT) region of osteopontin, an acid-rich, multidomain, phosphorylated 60kD glycoprotein. Osteopontin is a secreted chemokine-like protein that is a member of the SIBLING family, and is synthesised by many cell types.

Osteopontin is involved in enhancing production of interferon-gamma and interleukin-12 and reducing production of interleukin-10. It is essential in the pathway that leads to type I immunity. Osteoponitn is also thought to be important to cell-matrix interaction, and can bind fibronectin, collagen and calcium.

Osteopontin has been suggested to have a role in atherosclerosis, bone remodelling, angiogenesis, wound healing and tissue injuries.
Form/Format
Purified
Purified IgG - liquid, ready to use.
Concentration
IgG concentration 0.5mg/ml (varies by lot)
Sequence Length
300
Applicable Applications for anti-SPP1 antibody
ELISA (EIA), Western Blot (WB)
Application Notes
Western Blot: This item detects a band of approximately 60kDa in lysates of human thymus, tonsil and spleen. Approximate 30kDa band observed in lysates of cell lines Jurkat and MOLT4. This band corresponds to the cleavage product from the 60kDa glcoprotein.
ELISA: Maximum Dilution: 1/64000
Western Blotting: Minimum Dilution: 1.0; Maximum Dilution: 3.0 ug/ml
Perservative Stabilisers
0.02% Sodium Azide (NaN3)
0.5% Bovine Serum Albumin
Immunogen
Peptide with sequence C-DPKSKEEDKHLK, from the C-terminal of osteopontin.
Buffer Solution
TRIS buffered saline
Antiserum Preparation
Antisera to osteopontin (CT) were raised by repeated immunisations of goat with highly purified antigen. Purified IgG prepared by affinity chromatography.
Target Species
Human
Preparation and Storage
Store at 4 degree C or at -20 degree C if preferred. Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life: 18 months from date of despatch.

NCBI Description

The protein encoded by this gene is involved in the attachment of osteoclasts to the mineralized bone matrix. The encoded protein is secreted and binds hydroxyapatite with high affinity. The osteoclast vitronectin receptor is found in the cell membrane and may be involved in the binding to this protein. This protein is also a cytokine that upregulates expression of interferon-gamma and interleukin-12. Several transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2011]

Uniprot Description

osteopontin: Binds tightly to hydroxyapatite. Appears to form an integral part of the mineralized matrix. Probably important to cell-matrix interaction. Belongs to the osteopontin family. 5 isoforms of the human protein are produced by alternative splicing.

Protein type: Secreted; Secreted, signal peptide

Chromosomal Location of Human Ortholog: 4q22.1

Cellular Component: extracellular space; cell projection; perinuclear region of cytoplasm; apical part of cell; extracellular region

Molecular Function: protein binding; extracellular matrix binding; cytokine activity

Biological Process: neutrophil chemotaxis; extracellular matrix organization and biogenesis; decidualization; osteoblast differentiation; extracellular matrix disassembly; response to vitamin D; response to steroid hormone stimulus; biomineral formation; negative regulation of collateral sprouting of intact axon in response to injury; positive regulation of bone resorption; inflammatory response; cell adhesion; embryo implantation

Product Notes

The SPP1 spp1 (Catalog #AAA221360) is an Antibody produced from Goat and is intended for research purposes only. The product is available for immediate purchase. The GOAT ANTI HUMAN OSTEOPONTIN (C-TERMINAL) reacts with Dog (Expected from Sequence), Mouse (Expected from Sequence), Rat (Expected from Sequence) Antibody reactivity and working conditions may vary between species. and may cross-react with other species as described in the data sheet. AAA Biotech's OSTEOPONTIN can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Western Blot (WB). ELISA: Maximum Dilution: 1/64000 Western Blotting: Minimum Dilution: 1.0; Maximum Dilution: 3.0 ug/ml. Researchers should empirically determine the suitability of the SPP1 spp1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "OSTEOPONTIN, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
