Intended Uses: The kit is a sandwich enzyme immunoassay for in vitro quantitative measurement of MCSF in mouse serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

Principle of the Assay: The microplate provided in this kit has been pre-coated with an antibody specific to MCSF. Standards or samples are then added to the appropriate microplate wells with a biotin-conjugated antibody specific to MCSF. Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain MCSF, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of MCSF in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes a melanocyte-specific type I transmembrane glycoprotein. The encoded protein is enriched in melanosomes, which are the melanin-producing organelles in melanocytes, and plays an essential role in the structural organization of premelanosomes. This protein is involved in generating internal matrix fibers that define the transition from Stage I to Stage II melanosomes. This protein undergoes a complex pattern of prosttranslational processing and modification that is essential to the proper functioning of the protein. A secreted form of this protein that is released by proteolytic ectodomain shedding may be used as a melanoma-specific serum marker. Alternate splicing results in multiple transcript variants.
