Sensitivity
The minimum detectable dose of enkephalin is typically less than 3.56pg/mL. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined by subtracting two standard deviations to the mean optical density value of twenty zero standard replicates and calculating the corresponding concentration.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level enkephalin were tested 20 times on one plate, respectively. Intra-Assay: CV<10%
Inter-assay Precision

Intended Uses
The kit is a competitive inhibition enzyme immunoassay technique for the in vitro quantitative measurement of enkephalin in human serum, plasma and other biological fluids.
Preparation and Storage
For unopened kit, all the reagents should be kept according to the label on each vial. The Standard, Detection Reagent A, Detection Reagent B and the 96-well strip plate should be stored at -20 degree C while others should be at 4 degree C.

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. A monoclonal antibody specific to enkephalin has been pre-coated onto a microplate. A competitive inhibition reaction is launched between biotin labeled enkephalin and unlabeled enkephalin (Standards or samples) with the pre-coated antibody specific to enkephalin. After incubation the unbound conjugate is washed off. Next, avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The amount of bound HRP conjugate is reverse proportional to the concentration of enkephalin in the sample. After addition of the substrate solution, the intensity of color developed is reverse proportional to the concentration of enkephalin in the sample.

PENK: Met- and Leu-enkephalins compete with and mimic the effects of opiate drugs. They play a role in a number of physiologic functions, including pain perception and responses to stress. PENK(114-133) and PENK(237-258) increase glutamate release in the striatum. PENK(114-133) decreases GABA concentration in the striatum. Belongs to the opioid neuropeptide precursor family.
