Apoptosis is induced by certain cytokines including TNF and Fas ligand through their death domain-containing receptors. Several novel members in the TNF receptor family, including DR3, DR4, DR5 and DR6, function as cell death receptors. Two decoy receptors, DcR1 and DcR2, compete with DR4 and DR5 for binding of their ligand, TRAIL. Another decoy receptor, DcR3/TR6 was discovered recently. Unlike DcR1 and DcR2, DcR3 is a soluble rather than a membrane-associated molecule. DcR3 binds to FasL and LIGHT and inhibits FasLand LIGHT-induced apoptosis. DcR3 transcript is expressed in some lung and colon carcinomas and in some normal tissues.

NCBI Description

This gene belongs to the tumor necrosis factor receptor superfamily. The encoded protein is postulated to play a regulatory role in suppressing FasL- and LIGHT-mediated cell death. It acts as a decoy receptor that competes with death receptors for ligand binding. Over-expression of this gene has been noted in gastrointestinal tract tumors. Read-through transcription into this gene from the neighboring upstream gene, which encodes regulator of telomere elongation helicase 1 (RTEL1), generates a non-coding transcript.
