Principle of the Assay: The Picokine™ Human DCN Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Human DCN with a 96-well strip plate that is pre-coated with antibody specific for DCN. The detection antibody is a biotinylated antibody specific for DCN. The capture antibody is monoclonal antibody from mouse, the detection antibody is monoclonal antibody from mouse. The kit contains recombinant Human DCN with immunogen: Expression system for standard: Sf21; Immunogen sequence: G17-K359. The kit is analytically validated with ready to use reagents. To measure Human DCN, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density of the yellow product is linearly proportional to Human DCN in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human DCN in the sample.

Background/Introduction: Decorin is a proteoglycan on average 90 - 140 kilodaltons(kD) in size.It belongs to the small leucine-rich proteoglycan(SLRP) family and consists of a protein core containing leucine repeats with a glycosaminoglycan(GAG) chain consisting of either chondroitin sulfate(CS) or dermatan sulfate(DS).Using a genomic clone as the labeled probe and in situ hybridization of human metaphase chromosomes, decorin gene was mapped to the discrete region of human chromosome 12q23. Decorin is capable of suppressing the growth of various tumor cell lines when expressed ectopically.

NCBI Description

The protein encoded by this gene is a small cellular or pericellular matrix proteoglycan that is closely related in structure to biglycan protein. The encoded protein and biglycan are thought to be the result of a gene duplication. This protein is a component of connective tissue, binds to type I collagen fibrils, and plays a role in matrix assembly. It contains one attached glycosaminoglycan chain. This protein is capable of suppressing the growth of various tumor cell lines. There are multiple alternatively spliced transcript variants known for this gene. This gene is a candidate gene for Marfan syndrome. [provided by RefSeq, Jul 2008]

Corneal dystrophies are inherited, bilateral, primary alterations of the cornea that are not associated with prior inflammation or secondary to systemic disease. Most show autosomal dominant inheritance. Belongs to the small leucine-rich proteoglycan (SLRP) family. SLRP class I subfamily. 5 isoforms of the human protein are produced by alternative splicing.
