Western Blot (WB)
(Figure 1. Western blot analysis of SCRIBBLE using anti-SCRIBBLE antibody (MBS1750687). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human MCF-7 whole cell lysates,Lane 2: human COLO-320 whole cell lysates,Lane 3: human 22RV1 whole cell lysates,Lane 4: human SGC-7901 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-SCRIBBLE antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for SCRIBBLE at approximately 240KD. The expected band size for SCRIBBLE is at 175KD. )
Flow Cytometry (FC/FACS)
(Figure 2. Flow Cytometry analysis of A549 cells using anti-SCR1B antibody (MBS1750687).Overlay histogram showing A549 cells stained with MBS1750687 (Blue line). The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-SCR1B Antibody (MBS1750687,1ug/1x10^6 cells) for 30 min at 20 degree C. DyLight ®488 conjugated goat anti-rabbit IgG (5-10ug/1x10^6 cells) was used as secondary antibody for 30 minutes at 20 degree C. Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control. )

Protein Function: Scaffold protein involved in different aspects of polarized cells differentiation regulating epithelial and neuronal morphogenesis. Most probably functions in the establishment of apico-basal cell polarity. May function in cell proliferation regulating progression from G1 to S phase and as a positive regulator of apoptosis for instance during acinar morphogenesis of the mammary epithelium. May also function in cell migration and adhesion and hence regulate cell invasion through MAPK signaling. May play a role in exocytosis and in the targeting synaptic vesicles to synapses. Functions as an activator of Rac GTPase activity.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes a protein that was identified as being similar to the Drosophila scribble protein. The mammalian protein is involved in tumor suppression pathways. As a scaffold protein involved in cell polarization processes, this protein binds to many other proteins. The encoded protein binds to papillomavirus E6 protein via its PDZ domain and the C-terminus of E6. Two alternatively spliced transcript variants that encode different protein isoforms have been found for this gene. [provided by RefSeq, Nov 2011]

Product Notes
The SCRIBBLE scrib (Catalog #AAA1750687) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-SCRIBBLE Picoband antibody reacts with Human, Mouse, Rat No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's SCRIBBLE can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Flow Cytometry (FC/FACS), Immunohistochemistry (IHC), Immunocytochemistry (ICC), Western Blot (WB). WB: 0.1-0.5mug/ml Immunohistochemistry(Frozen Section): 0.5-1mug/ml ICC: 0.5-1mug/ml FC/FACS: 1-3g/1x10 6 cells Direct ELISA: 0.1-0.5mug/ml. Researchers should empirically determine the suitability of the SCRIBBLE scrib for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SCRIBBLE, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
