Western Blot (WB)
(Figure 1. Western blot analysis of Sacsin using anti-Sacsin antibody (MBS1750427).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions.Lane 1: rat brain tissue lysates,Lane 2: mouse brain tissue lysates,Lane 3: human Hela cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Sacsin antigen affinity purified polyclonal antibody at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for Sacsin at approximately 521KD. The expected band size for Sacsin is at 521KD. )
Immunohistochemistry (IHC)
(Figure 2. IHC analysis of Sacsin using anti-Sacsin antibody (MBS1750427).Sacsin was detected in paraffin-embedded section of mouse brain tissue. Heat mediated antigen retrieval was performed in citrate buffer (pH6, epitope retrieval solution) for 20 mins. The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1ug/ml rabbit anti-Sacsin Antibody (MBS1750427) overnight at 4 degree C. Biotinylated goat anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37 degree C. The tissue section was developed using Strepavidin-Biotin-Complex (SABC) with DAB as the chromogen. )

Protein Function: Co-chaperone which acts as a regulator of the Hsp70 chaperone machinery and may be involved in the processing of other ataxia-linked proteins.
NCBI and Uniprot Product Information
NCBI Description
This gene encodes the sacsin protein, which includes a UbL domain at the N-terminus, a DnaJ domain, and a HEPN domain at the C-terminus. The gene is highly expressed in the central nervous system, also found in skin, skeletal muscles and at low levels in the pancreas. This gene includes a very large exon spanning more than 12.8 kb. Mutations in this gene result in autosomal recessive spastic ataxia of Charlevoix-Saguenay (ARSACS), a neurodegenerative disorder characterized by early-onset cerebellar ataxia with spasticity and peripheral neuropathy. The authors of a publication on the effects of siRNA-mediated sacsin knockdown concluded that sacsin protects against mutant ataxin-1 and suggest that "the large multi-domain sacsin protein is able to recruit Hsp70 chaperone action and has the potential to regulate the effects of other ataxia proteins" (Parfitt et al., PubMed: 19208651). A pseudogene associated with this gene is located on chromosome 11. Alternative splicing of this gene results in multiple transcript variants. [provided by RefSeq, May 2013]

The SACS sacs (Catalog #AAA1750427) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-Sacsin Picoband antibody reacts with Human, Mouse, Rat No cross reactivity with other proteins. and may cross-react with other species as described in the data sheet. AAA Biotech's Sacsin can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA), Immunohistochemistry (IHC), Western Blot (WB). WB: Concentration: 0.1-0.5ug/ml IHC-P: Concentration: 0.1-1ug/ml; Antigen Retrieval: By Heat ELISA: Concentration: 0.1-0.5ug/ml Tested Species: In-house tested species with positive results. Enhanced Chemiluminescent Kit with anti-rabbit IgG (MBS176460) for western blot and HRP conjugated anti-rabbit IgG super vision assay kit (Please Inquire) for IHC-P. Researchers should empirically determine the suitability of the SACS sacs for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Sacsin, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
