Host
Baculovirus Sf9 Cells
Purity/Purification
Greater than 95.0% as determined by: (a) Analysis by RP-HPLC. (b) Analysis by SDS-PAGE.
Form/Format
The protein was lyophilized with no additives.
Sterile Filtered White lyophilized (freeze-dried) powder.
Sequence
The sequence of the first five N-terminal amino acids was determined and was found to be Thr-Gln-Asp-Cys-Ser
Sequence Length
235
Solubility
It is recommended to reconstitute the lyophilized Flt3-Ligand in sterile 18M Omega -cm H2O not less than 100 ug/ml, which can then be further diluted to other aqueous solutions.
Protein Content
Protein quantitation was carried out by two independent methods:1. UV spectroscopy at 280 nm using the absorbency value of 1.42 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a standard of Flt3-Ligand as a Reference Standard.
Biological Activity
The ED50, calculated by the dose-dependent stimulation of the proliferation of human MUTZ-2 cells is < 3.0 ng/ml.
Preparation and Storage
Lyophilized Flt3-Ligand although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution Flt3-L should be stored at 4 degree C between 2-7 days and for future use below -18 degree C.For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).Please prevent freeze-thaw cycles.

NCBI Description

Dendritic cells (DCs) provide the key link between innate and adaptive immunity by recognizing pathogens and priming pathogen-specific immune responses. FLT3LG controls the development of DCs and is particularly important for plasmacytoid DCs and CD8 (see MIM 186910)-positive classical DCs and their CD103 (ITGAE; MIM 604682)-positive tissue counterparts (summary by Sathaliyawala et al., 2010 [PubMed 20933441]).[supplied by OMIM, Jan 2011]

Uniprot Description

FLT3LG: Stimulates the proliferation of early hematopoietic cells by activating FLT3. Synergizes well with a number of other colony stimulating factors and interleukins. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Cytokine; Ligand, receptor tyrosine kinase

Chromosomal Location of Human Ortholog: 19q13.3

Cellular Component: extracellular space; membrane; integral to membrane

Molecular Function: protein homodimerization activity; cytokine activity; receptor tyrosine kinase binding; receptor binding

Biological Process: positive regulation of natural killer cell differentiation; regulation of myeloid dendritic cell activation; lymphocyte differentiation; embryonic hemopoiesis; positive regulation of cell proliferation; positive regulation of protein amino acid phosphorylation; signal transduction; positive regulation of myoblast differentiation

Product Notes

The FLT3LG flt3lg (Catalog #AAA142285) is an Active Protein produced from Baculovirus Sf9 Cells and is intended for research purposes only. The product is available for immediate purchase. The amino acid sequence is listed below: The sequence of the first five N-terminal amino acids was determined and was found to be Thr-Gln-As p-Cys-Ser. It is sometimes possible for the material contained within the vial of "Flt3-Ligand, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
