Mouse Prekallikrein Total Antigen Assay ELISA kit
Principle of the Assay: Mouse prekallikrein will bind to t he monoclonal capture antibody coated on the micrptiter plate. Preka llikrein and kallikrein will react with the antibody on the plate. After appropriate washing steps, anti-mouse prekallikrein primary antibody binds to the captured protein. Excess primary antibody is washed away and bound antibody is reacted with horseradish peroxidase conjugated streptavidin. Following an additional washing step, TM B substrate is used for color development at 450nm. A standard calibration curve is prepared along with the samples to be measured using dilutions of mouse prekallikrein. Color development is proportional to the concentration of total prekallikrein in the samples!!Background/Introduction: Preka llikrein is the glycosylated single chain zymogen precursor of the plasma serine protease kall ikrein. Plasma prekallikrein circulates with kininogen and is activated by Factor Xlla in the intrinsic coagulation pathway. Kallikrein activates plasminogen in fibrinolysis and cleaves kininogen in the bradykinin system of vasodilation. Prekallikrein deficiency is rare and causes increased activated partial thromboplastin time [1]. Elevated plasma prekallikrein is associated with diabetes [2] and cardiovascular disease [3].

This gene encodes a member of the kallikrein subfamily of serine proteases that are involved in diverse physiological functions such as skin desquamation, tooth enamel formation, seminal liquefaction, synaptic neural plasticity and brain function. The encoded preproprotein undergoes proteolytic processing to generate a disulfide-linked heterodimeric enzyme comprised of heavy and light chains. A complete deletion of the encoded protein prevents occlusive thrombus formation in mice with a minimal role in provoked bleeding. [provided by RefSeq, May 2016]

The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin.
