A zinc finger protein is a DNA-binding protein domain consisting of zinc fingers ranging from two in the Drosophila regulator ADR1, the more common three in mammalian Sp1 up to nine in TFIIIA. They occur in nature as the part of transcription factors conferring DNA sequence specificity as the DNA-binding domain.
By EST database analysis, followed by PCR of a human B lymphoblastoid cell line cDNA library, Herberg et al. (1998) cloned ZBTB22, which they called BING1. The deduced 634-amino acid protein contains an N-terminal POZ/BTB domain and a C-terminal C2H2-type zinc finger domain.By genomic sequence analysis, Herberg et al. (1998) mapped the ZBTB22 gene to a gene cluster on chromosome 6p21.3 that includes tapasin .
Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ZBTB22 in samples. An antibody specific for ZBTB22 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyZBTB22 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ZBTB22 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ZBTB22 bound in the initial step. The color development is stopped and the intensity of the color is measured.
