Intended Uses: This VIM ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine VIM. This ELISA kit for research use only!

Principle of the Assay: VIM ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-VIM antibody and an VIM-HRP conjugate. The assay sample and buffer are incubated together with VIM-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the VIM concentration since VIM from samples and VIM-HRP conjugate compete for the anti-VIM antibody binding site. Since the number of sites is limited, as more sites are occupied by VIM from the sample, fewer sites are left to bind VIM-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The VIM concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the intermediate filament family. Intermediate filamentents, along with microtubules and actin microfilaments, make up the cytoskeleton. The protein encoded by this gene is responsible for maintaining cell shape, integrity of the cytoplasm, and stabilizing cytoskeletal interactions. It is also involved in the immune response, and controls the transport of low-density lipoprotein (LDL)-derived cholesterol from a lysosome to the site of esterification. It functions as an organizer of a number of critical proteins involved in attachment, migration, and cell signaling. Mutations in this gene causes a dominant, pulverulent cataract.[provided by RefSeq, Jun 2009]
