Principle of the Assay: USP6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-USP6 antibody and an USP6-HRP conjugate. The assay sample and buffer are incubated together with USP6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the USP6 concentration since USP6 from samples and USP6-HRP conjugate compete for the anti-USP6 antibody binding site. Since the number of sites is limited, as more sites are occupied by USP6 from the sample, fewer sites are left to bind USP6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The USP6 concentration in each sample is interpolated from this standard curve.

Uniprot Description

USP6: Deubiquitinase with an ATP-independent isopeptidase activity, cleaving at the C-terminus of the ubiquitin moiety. Catalyzes its own deubiquitination. In vitro, isoform 2, but not isoform 3, shows deubiquitinating activity. Promotes plasma membrane localization of ARF6 and selectively regulates ARF6- dependent endocytic protein trafficking. Is able to initiate tumorigenesis by inducing the production of matrix metalloproteinases following NF-kappa-B activation. A chromosomal aberration involving USP6 is a common genetic feature of aneurysmal bone cyst, a benign osseous neoplasm. Translocation t(16;17)(q22;p13) with CDH11. The translocation generates a fusion gene in which the strong CDH11 promoter is fused to the entire USP6 coding sequence, resulting in USP6 transcriptional up-regulation. Belongs to the peptidase C19 family. 3 isoforms of the human protein are produced by alternative splicing.
