Principle of the Assay: ULBP4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ULBP4 antibody and an ULBP4-HRP conjugate. The assay sample and buffer are incubated together with ULBP4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ULBP4 concentration since ULBP4 from samples and ULBP4-HRP conjugate compete for the anti-ULBP4 antibody binding site. Since the number of sites is limited, as more sites are occupied by ULBP4 from the sample, fewer sites are left to bind ULBP4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ULBP4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene belong to the RAET1 family, which consists of major histocompatibility complex (MHC) class I-related genes located in a cluster on chromosome 6q24.2-q25.3. This and RAET1G protein differ from other RAET1 proteins in that they have type I membrane-spanning sequences at their C termini rather than glycosylphosphatidylinositol anchor sequences. This protein functions as a ligand for NKG2D receptor, which is expressed on the surface of several types of immune cells, and is involved in innate and adaptive immune responses. Alternatively spliced transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Aug 2011]
