Principle of the Assay: UINS ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-UINS antibody and an UINS-HRP conjugate. The assay sample and buffer are incubated together with UINS-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the UINS concentration since UINS from samples and UINS-HRP conjugate compete for the anti-UINS antibody binding site. Since the number of sites is limited, as more sites are occupied by UINS from the sample, fewer sites are left to bind UINS-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The UINS concentration in each sample is interpolated from this standard curve.

ULBP1: Ligand for the NKG2D receptor, together with at least ULBP2 and ULBP3. ULBPs activate multiple signaling pathways in primary NK cells, resulting in the production of cytokines and chemokines. Binding of ULBPs ligands to NKG2D induces calcium mobilization and activation of the JAK2, STAT5, ERK and PI3K kinase/Akt signal transduction pathway. In CMV infected cells, interacts with soluble CMV glycoprotein UL16. The interaction with UL16 blocked the interaction with the NKG2D receptor, providing a mechanism by which CMV infected cells might escape the immune system. UL16 also causes ULBP1 to be retained in the ER and cis- Golgi apparatus so that it does not reach the cell surface. Belongs to the MHC class I family.
