Scientific Background: UBE2E3 also known as UBCH9 or ubiquitin-conjugating enzyme E2E 3 is a member of the E2 ubiquitin-conjugating enzyme family. Ubiquitination, the covalent attachment of the protein ubiquitin (Ub) to other cellular proteins is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation that involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s(1). UBE2E3 is highly expressed in skeletal muscle (2). UBE2E3 is essential for the proliferation of RPE-1 cells and is downregulated during RPE layer maturation in the developing mouse eye. These findings indicate that UBE2E3 is a major enzyme in modulating the balance between RPE cell proliferation and differentiation.

NCBI and Uniprot Product Information
NCBI Description
The modification of proteins with ubiquitin is an important cellular mechanism for targeting abnormal or short-lived proteins for degradation. Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes, or E1s, ubiquitin-conjugating enzymes, or E2s, and ubiquitin-protein ligases, or E3s. This gene encodes a member of the E2 ubiquitin-conjugating enzyme family. The encoded protein shares 100% sequence identity with the mouse and rat counterparts, which indicates that this enzyme is highly conserved in eukaryotes. Multiple alternatively spliced transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jun 2013]
Uniprot Description
Function: Accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro catalyzes 'Lys-11'- and 'Lys-48'-, as well as 'Lys-63'-linked polyubiquitination. Participates in the regulation of transepithelial sodium transport in renal cells. May be involved in cell growth arrest. Ref.1 Ref.11
Catalytic activity: ATP + ubiquitin + protein lysine = AMP + diphosphate + protein N-ubiquityllysine.
Pathway: Protein modification; protein ubiquitination.
Subunit structure: The ubiquitin-loaded form interacts specifically with importin-11 (IPO11), leading to its import into the nucleus
By similarity. Interacts with NEDD4L.
Subcellular location: Nucleus. Cytoplasm. Note: Shuttles between the nucleus and cytoplasm in a IPO11-dependent manner. Ref.9
Tissue specificity: Ubiquitously expressed at low levels. Highly expressed in skeletal muscle. Ref.1
Sequence similarities: Belongs to the ubiquitin-conjugating enzyme family.

The tag for this protein is His tag!!Expression System||E.coli!!Source Note||Recombinant full length human UBE2E3 (UBCH9) was expressed in E. coli cells. AAA Biotech's UBE2E3 (UBCH9) can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the UBE2E3 (UBCH9) ube2e3 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "UBE2E3 (UBCH9), Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
