Intended Uses: This UBC ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human UBC. This ELISA kit for research use only!

Principle of the Assay: UBC ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-UBC antibody and an UBC-HRP conjugate. The assay sample and buffer are incubated together with UBC-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the UBC concentration since UBC from samples and UBC-HRP conjugate compete for the anti-UBC antibody binding site. Since the number of sites is limited, as more sites are occupied by UBC from the sample, fewer sites are left to bind UBC-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The UBC concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene represents a ubiquitin gene, ubiquitin C. The encoded protein is a polyubiquitin precursor. Conjugation of ubiquitin monomers or polymers can lead to various effects within a cell, depending on the residues to which ubiquitin is conjugated. Ubiquitination has been associated with protein degradation, DNA repair, cell cycle regulation, kinase modification, endocytosis, and regulation of other cell signaling pathways. ubiquitin: a peptide 76 amino acids in length that can be covalently attached to target lysines either as a monomer or as a lysine-linked polymer. Ubiquitin is encoded by 4 different genes. UBA52 and RPS27A genes code for a single copy of ubiquitin fused to the ribosomal proteins L40 and S27a, respectively. UBB and UBC genes code for a polyubiquitin precursor with exact head to tail repeats, the number of repeats differ between species and strains. Only the 76 amino acids of monoubiquitin product are shown in this entry. At the protein level, it is not possible to determine which of the four genes a given ubiquitin chain was derived from. Hundreds of ubiquitin ligases and hydrolases have been identified, implicating ubiquitin as a major regulatory element in many crucial cellular systems. It can be covalently bound to target proteins via an isopeptide bond either as a monomer (monoubiquitin), a polymer linked via different Lys residues of the ubiquitin (polyubiquitin chains) or a linear polymer linked via the initiator Met of the ubiquitin (linear polyubiquitin chains). Polyubiquitin chains, when attached to a target protein, have different functions depending on the Lys residue of the ubiquitin that is linked: Lys-6-linked may be involved in DNA repair; Lys-11-linked is involved in ERAD (endoplasmic reticulum-associated degradation) and in cell-cycle regulation; Lys-29-linked is involved in lysosomal degradation; Lys-33-linked is involved in kinase modification; Lys-48-linked is involved in protein degradation via the proteasome; Lys-63-linked is involved in endocytosis, DNA-damage responses as well as in signaling processes leading to activation of the transcription factor NF-kappa-B. Linear polymer chains formed via attachment by the initiator Met lead to cell signaling. Ubiquitin is usually conjugated to Lys residues of target proteins, however, in rare cases, conjugation to Cys or Ser residues has been observed. When polyubiquitin is free (unanchored-polyubiquitin), it also has distinct roles, such as in activation of protein kinases, and in signaling.
