Principle of the Assay: u-PAR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-u-PAR antibody and an u-PAR-HRP conjugate. The assay sample and buffer are incubated together with u-PAR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the u-PAR concentration since u-PAR from samples and u-PAR-HRP conjugate compete for the anti-u-PAR antibody binding site. Since the number of sites is limited, as more sites are occupied by u-PAR from the sample, fewer sites are left to bind u-PAR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The u-PAR concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes the receptor for urokinase plasminogen activator and, given its role in localizing and promoting plasmin formation, likely influences many normal and pathological processes related to cell-surface plasminogen activation and localized degradation of the extracellular matrix. It binds both the proprotein and mature forms of urokinase plasminogen activator and permits the activation of the receptor-bound pro-enzyme by plasmin. The protein lacks transmembrane or cytoplasmic domains and may be anchored to the plasma membrane by a glycosyl-phosphatidylinositol (GPI) moiety following cleavage of the nascent polypeptide near its carboxy-terminus. However, a soluble protein is also produced in some cell types. Alternative splicing results in multiple transcript variants encoding different isoforms. The proprotein experiences several post-translational cleavage reactions that have not yet been fully defined. [provided by RefSeq, Jul 2008]
