Uniprot Description

TTC3: an E3 ubiquitin ligase that mediates the ubiquitination and subsequent degradation of phosphorylated Akt in the nucleus, acting to dampen Akt signaling after its activation. The human protein is phosphorylated on S378, a prerequisite for ubiquitin ligase activity. The kinase responsible for this phosphorylation may be Akt, suggestive of the existence of an auto-feedback loop that degrades Akts levels after it is activated. However the pattern of amino acids surrounding S378 only partially matches the canonical Akt motif, suggesting that kinase other than Akt may be responsible for the phosphorylation at S378. Catalyzes the formation of K48-polyubiquitin chains. May play a role in neuronal differentiation inhibition via its interaction with CRIK. TTC3 is one of the supernumerary genes in the Down?s syndrome critical region. Three alternatively spliced human isoforms of have been reported.
