This assay has high sensitivity and excellent specificity for detection of TSPO. No significant cross-reactivity or interference between TSPO and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between TSPO and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This TSPO ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey TSPO. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TSPO ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-TSPO antibody and an TSPO-HRP conjugate. The assay sample and buffer are incubated together with TSPO-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TSPO concentration since TSPO from samples and TSPO-HRP conjugate compete for the anti-TSPO antibody binding site. Since the number of sites is limited, as more sites are occupied by TSPO from the sample, fewer sites are left to bind TSPO-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TSPO concentration in each sample is interpolated from this standard curve.

NCBI Description

Present mainly in the mitochondrial compartment of peripheral tissues, the protein encoded by this gene interacts with some benzodiazepines and has different affinities than its endogenous counterpart. The protein is a key factor in the flow of cholesterol into mitochondria to permit the initiation of steroid hormone synthesis. Alternatively spliced transcript variants have been reported; one of the variants lacks an internal exon and is considered non-coding, and the other variants encode the same protein.
